Early CTE disease process found to be mechanistically different than what occurs in late stages BUMedicine
Our understanding of the molecular underpinnings of chronic traumatic encephalopathy and its associated pathology in post-mortem brain is incomplete. Factors including years of play and genetic risk variants influence the extent of tau pathology associated with disease expression, but how these factors affect gene expression, and whether those effects are consistent across the development of disease, is unknown.
The primary risk factor for developing CTE is the duration of exposure to repetitive head impacts , which can be measured as total years of play for contact sport athletes []. The neurological symptoms of CTE often only manifest decades after players have retired, suggesting the effects of RHI set in motion a progression of pathological events ultimately leading to disease.
Our previous work showed that two genetic risk variants in the Apolipoprotein E and Transmembrane Protein 106B genes also influence the frequency and severity of CTE. The association of the
