We assessed the RNA quality with Agilent Bioanalyzer 2100 using an RNA 6000 Pico Kit , and detected RNA quality with Nanodrop 1000 . Only RNAs with high quality were used for cDNA synthesis and amplification following the protocol as previously described. Then, the primitive sequencing libraries were prepared and individually barcoded using the Nextera XT DNA Library Prep Kit following the manufacturer’s protocol.
5% PEG‐6000/NaCl/Tris and 10 μl prepared Dynabeads were added to the supernatant and mixed . The mixture was incubated for 5 min followed by bead separation on a magnetic stand. This time, the supernatant was discarded, and the beads were collected. The beads were washed twice with 70% ethanol and dried for 5 min. The tubes were then taken off the magnetic stand, and DNA was eluted from the beads by resuspending them in 15 μl EB.
