We're super excited about the release of your new album. What can you tell us about your new music?I did a couple of albums when I was a lot younger, and this is the first album I've written, produced, sung, and played. The album is calledbecause the industry's been an absolute, as you know... how do I say it without swearing? Challenging! Let's say it's been challenging.

I'm a really strong woman, and I really want to put exactly what I hear in my head out there. So, it took some time to actually be able to do that, you know? It's a testament to how great the album is that I did it, and I'm so proud of it. Just the fact of where I've been to make it, that to me is the victory. Like what else it [the album] does, what people think of it, and how many streams it gets, it doesn't matter and has absolutely no consequence.

I mean, when I started, there wasn't streaming and all that stuff. It has completely changed, but I don't think the gatekeepers have, if that makes sense. It's still very misogynistic. It's still a boys club, you know? If you're a 16-year-old girl, you know, that's great, but if you're a woman, you have no value to them, so in that way, it's really misogynistic. It's about the choices that they make and the money that they put behind people.

Biomarkers for early prediction of therapy success are missing. We investigated the dynamics of gene expression and DNA methylation in blood samples of IBD patients treated with the TNF antagonist infliximab and analyzed the predictive potential regarding therapy outcome. Methods We performed a longitudinal, blood-based multi-omics study in two prospective IBD patient cohorts receiving first-time infliximab therapy (discovery: 14 patients, replication: 23 patients). Samples were collected at up to 7 time points (from baseline to 14 weeks after therapy induction). RNA-sequencing and genome-wide DNA methylation data were analyzed and correlated with clinical remission at week 14 as a primary endpoint. Results We found no consistent ex ante predictive signature across the two cohorts. Longitudinally upregulated transcripts in the non-remitter group comprised TH2- and eosinophil-related genes including ALOX15, FCER1A, and OLIG2. Network construction identified transcript modules that were coherently expressed at baseline and in non-remitting patients but were disrupted at early time points in remitting patients. These modules reflected processes such as interferon signaling, erythropoiesis, and platelet aggregation. DNA methylation analysis identified remission-specific temporal changes, which partially overlapped with transcriptomic signals. Machine learning approaches identified features from differentially expressed genes cis-linked to DNA methylation changes at week 2 as a robust predictor of therapy outcome at week 14, which was validated in a publicly available dataset of 20 infliximab-treated CD patients. Single-cell RNAseq uncovers cellular heterogeneity and provides a signature for pediatric sleep apneaBackground Obstructive Sleep Apnea (OSA) is a highly prevalent disease and a major cause of systemic inflammation leading to neurocognitive, behavioral, metabolic, and cardiovascular dysfunction in children and adults. However, the impact of OSA on the heterogeneity of circulating immune cells remains to be determined. Methods We applied single-cell transcriptomics analysis (scRNA-seq) to identify OSA-induced changes in transcriptional landscape in PBMC composition which uncovered severity-dependent differences in several cell lineages. Furthermore, a machine learning approach was used to combine scRNAs-seq cell-specific markers with those differentially expressed in OSA. Results scRNA-seq demonstrated OSA-induced heterogeneity in cellular composition and enabled the identification of previously undescribed cell types in PBMCs. We identified a molecular signature consisting of 32 genes, which distinguished OSA patients from various controls with high precision (AUC=0.96), and accuracy (93% PPV and 95% NPV, respectively) in an independent PBMC bulk RNA expression dataset. Conclusion OSA deregulates systemic immune function and displays a molecular signature that can be assessed in standard cellular RNA without the need for pre-analytical cell separation, thereby making the assay amenable to application in a molecular diagnostic setting.
